چکیده

Previous studies have suggested a "catalytic role" in neoplastic angiogenesis and cancer progression for bone marrow–derived endothelial progenitor cells (EPC). However, preclinical and clinical studies have shown that the quantitative role of marrow-derived EPCs in cancer vascularization is extremely variable. We have found that human and murine white adipose tissue (WAT) is a very rich reservoir of CD45-CD34þ EPCs with endothelial differentiation potential, containing a mean of 263 times more CD45-CD34þ cells/mL than bone marrow. Compared with marrow-derived CD34þ cells mobilized in blood by granulocyte colony–stimulating factor, purified WAT-CD34þ cells expressed similar levels of stemness-related genes, significantly increased levels of angiogenesis-related genes, and increased levels of FAP-a, a crucial suppressor of antitumor immunity. In vitro, WAT-CD34þ cells generated mature endothelial cells and capillary tubes as efficiently as mature mesenchymal cells. The coinjection of human WAT-CD34þ cells from lipotransfer procedures contributed to tumor vascularization and significantly increased tumor growth andmetastases in several orthotopicmodels of human breast cancer in immunodeficient mice. Endothelial cells derived from human WAT-CD34þ cells lined the lumen of cancer vessels. These data indicate that CD34þ WAT cells can promote cancer progression and metastases. Our results highlight the importance of gaining a better understanding of the role of different WAT-derived cells used in lipotransfer for breast reconstruction in patients with breast cancer. STAT3 as a Key Factor in Tumor Microenvironment and Cancer Stem Cell

Background Recent studies revealed that tumor-associated macrophages (TAMs) play a decisive role in the regulation of tumor progression by manipulating tumor oncogenesis, angiogenesis and immune functions within tumor microenvironments. Signal transducer and activator of transcription 3 (STAT3), which is a point of convergence for numerous oncogenic signalling pathways, is constitutively activ...

Histological Evidences after Platelet-Rich-Plasma and Adipose Drived Stem Cells Injection on Critical Size Cleft Palate

Background Cleft palate (CP) is a common congenital defect. It makes serious difficulties for cleft-affected children. The gold standard of care is autogenous bone grafting which may cause additional problems in donor site along with disappointing results. Tissue engineering is a promising solution for a widespread range of defects and disorders. It is reasonable to utilize this novel technolo...

Biological and Clinicopathological Significance of Cripto-1 Expression in the Progression of Human ESCC

Background: Human Cripto-1, a member of the EGF-CFC family, is involved in embryonic development, embryonic stem cell maintenance, and tumor progression. It also participates in multiple cell signaling pathways including Wnt, Notch, and TGF-β. Remarkably, it is expressed in cancer stem cell (CSC) compartments, boosting tumor cell migration, invasion, and angiogenesis. Although Cripto-1 is ...

Mesenchymal Stem Cells Derived from Rat Epicardial Versus Epididymal Adipose Tissue

Objective(s) Some investigation has indicated that adipose-derived stem cells possess different surface epitopes and differentiation potential according to the localization of fat pad from which the cells were derived. In the present study proliferation capacity and aging of such cells were explored. Materials and Methods Adherent cells were isolated from the collagenase digests of adipose tiss...
